It is produced by an enzyme, prostacyclin endoperoxidase from cyclic endoperoxides in endothelium cells [7].

The endothelium represents a highly active autocrine, paracrine and endocrine organ synthetizing many vasoactive substances, including vasoconstrictors (thromboxane A2, endothelin-1, prostaglandin H2) and vasodilators such as nitric oxide (NO) [65], prostacyclin (PGI2) [66,67] and endothelium-derived hyperpolarizing factors (EDHF) [68]. The relative contribution of dilating and constricting substances to maintain vascular tone and adequate tissue perfusion during both rest and exercise depends on the vessel diameter and vascular bed.

In 1976 Richard Gryglewski, Stuart Bunting, Salvador Moncada, Rod Flower and others discovered and characterised a novel prostaglandin produced from vascular tissue with striking biological properties [37], [30]). This compound, originally known as ‘prostaglandin X’ was subsequently identified and synthesised in conjunction with the Upjohn company and subsequently named prostacyclin [31].
